The immunological response to a xenograft is strong and difficult to suppress. It is driven both by the humoral and cellular part of the immune system. The aim of this thesis was to characterise and modulate this response in a concordant mouse-to-rat model, using both vascularised and non-vascularised grafts. Exposure of mouse cells or tissue to the circulation of a rat, either through transplantation or transfusions, easily evoked an immune response, consisting of IgM antibodies. A response that was aimed both at antigens present on mouse mononuclear cells and on erythrocytes. A non-immunosuppressed rat rejected a mouse heart graft within three days. The combined use of cyclosporine A (CyA) and deoxyspergualin (DSG) as immunosuppression prevented the rejection of vascularised heart transplants as well as of non-vascularised pancreatic islet grafts. This acceptance was sustained for the heart transplant also after the termination of DSG treatment, but not for the pancreatic islet graft. Furthermore, a second heart graft was accepted when transplanted under monotherapy with CyA 56-154 days after the first transplantation. This finding was interpreted as a humoral unresponsiveness, which could not be reproduced when the primary heart was substituted with a cellular graft, consisting of pancreatic islets or heart cells, or by blood transfusions. However, the rejection of a mouse heart after blood transfusions occurred in the absence of antibodies directed against mouse erythrocytes, in contrast to the observations in non-transfused animals. This indicates that a partial humoral tolerance restricted to the response against erythrocytes can be induced. This mechanism may offer a possibility to induce total humoral tolerance against a xenograft if the appropriate antigens are administered in conjunction with CyA and DSG.

P-45: Xenotransplantation of Mouse Ovaries into Pseudopregnant Rat Uteri
Background Xenotransplantation of ovary can help individuals who may be at sterility risk after chemotherapy or radiotherapy of diseases such as cancer. Moreover, this technique can be used to preserve fertility of valuable animal species. The aim of the present study was to preserve balb/c mice ovarian tissue by xenotransplantation into uterus of rat that is immune suppressed. MaterialsAndMeth...
متن کاملCan Allergic Disorders Decrease the Risk of Thromboembolic Events in Atherosclerosis? an Evidence-based Review
Atherosclerosis is a complex inflammatory disorder of the medium and large-sized arteries. Recent experiments demonstrated that atherosclerosis is a Th1 dominant autoimmune condition, whereas Th2 cells are rarely detected within the atherosclerotic lesions. Several studies have indicated that Th2 type cytokines could be effective in the reduction and stabilization of atherosclerotic plaque. The...
متن کاملModulation Response and Relative Intensity Noise Spectra in Quantum Cascade Lasers
Static properties, relatively intensity noise and intensity modulation response in quantum cascade lasers (QCLs) studied theoretically in this paper. The present rate equations model consists of three equations for the electrons density in the conduction band and one equation for photons density in cavity length. Two equations were derived to calculate the noise and modulation response. Calcula...
متن کاملPhysiological and Pathological Roles for MicroRNAs: Implications for Immunity Complications
MicroRNAs (miRNAs) are small non-coding regulatory RNAs molecules with a size of approximately 22 nucleotides that are implicated in regulating gene expression at the post-transcriptional regulatory levels. Inflammatory disorders especially autoimmune diseases (ADs) occur from an abnormal immune response of body against cells of their own specific tissues or multiple organ systems leading to ch...
متن کاملPhysiologic and immunologic hurdles to xenotransplantation.
The major problem in the field of renal transplantation is currently the shortage of available kidneys. However, the use of animals as a source of kidneys, i.e., xenotransplantation, is increasingly being viewed as a potential solution to this problem. One preeminent hurdle to xenotransplantation is the immune response of the recipient against the graft; other hurdles include the physiologic li...
